My Emirates flight should land at Terminal 3 at 6:10 AM. My connecting flight from terminal 2 is scheduled for 10:10 AM.
What is the faster and most reliable way to transfer between the two terminals?
Grab or taxi.
Or do like most on here and over night in Manila between flights and take the risk and stress out of the situation.
Your flight is on the borderline on my risk levels. Any significant flight delay means you miss your next flight.
Assuming your inbound is on time and you process through NAIA in a reasonable fashion (big*IF*) you will be hitting the weekday Manila rush when you eventually emerge from T3. T2 is the other side of the airport to T3 and several Kms away and the only way to go between them is on public roads, Manila is one of the most congested city's on the planet, see where I'm going with this? Coming back to that big *IF*, NAIA is also not an efficient airport before now among other things I have sat on an aeroplane on the apron for half a hour waiting for an air bridge to come available, some have report a lot longer. Also, don't forget check-in/bag drop for your onward flight closes a hour before departure, so you automatically lose a hour.
Its a roll of the dice if you will make your onward flight or not, I for one will not be rushing down the bookies and putting money on you making it. :-)

More specifically, We will land on Sunday morning and don't have the flexibility to modify our connection flight. What is the fastest way to transfer between the terminals?
Sunday morning is a good time
There is an inter terminal bus but if you are told there is a wait of more than half an hour get a taxi from the line outside the terminal but if you find you only have 2.5 hours or less until your onward flight departs after clearing customs get a taxi

I don't know why people put themselves through things like this? A quick glance at the forum identifies the not inconsequential and inconsiderable risks of short transfer times at Manila airport.
.......or they ask the question after they already booked a short connection.
The quickest would be Grab or taxi - i would choose Grab. Even on a weekend there can be a lot of traffic.
What gets me, even a self connect of this nature at an efficient European and Middle East airport with connected terminals would not be without risk, never mind undertaking at it NAIA with all its shortcomings, which is far from a state secret. Not having flexibility could come at quite a price if things don't work out.
