Car rental pick up airport or downtown

My husband and I are flying into Boston end of April next year. We are in Boston for a couple of nights staying at the Boston marina. We then pick up a rental car to drive to Cape Cod and our tour of New England for a couple of weeks.

The closest rental to our hotel is in Atlantic Ave so does it make sense to pick up from there or go back to the airport? We will drop off at the airport as we will be flying out of Boston. It does seem cheaper to hire from the airport.

Which pick up is better for driving out of Boston on a Thursday to Falmouth. When does rush our finish?
